Design Notes

We inserted PvuII recognition domains to the two ends of the gate by Golden Gate assembly. The insertion of the sequence to Psb1C3 was performed through the Golden Gate assembly by our modified AmilCP construct (part: ). The scar produced through the assembly produces both PvuII sites necessary for our gate to function.
